As a Scientist in the field of Molecular Biology and Genetic Engineering/ Cell Line Production, you will play a crucial role in the development of tools and assays requiring introduction of genetic changes of various nature (e.g. gene knock-out, silencing, gene overexpression). The developed tools and assays will be used in the Drug Discovery process and products in the GMP environment for biotech and pharma companies from Europe, USA and the rest of the world.

Key Responsibilities
  • Molecular cloning (design and conduct of the cloning – e.g. bacteria transformation, plasmid amplification and isolation, colony PCR).
  • Genome engineering – design and cloning of gRNA.
  • Production of lentiviral vectors.
  • Culture of eukaryotic cell lines and cell line banking.
  • Introduction of RNA/DNA/protein into the eukaryotic cell lines (chemical transfection, electroporation, transduction with viral vectors).
  • Gene silencing through siRNA.
  • Designing of primers for PCR and qPCR.
  • Conducting PCR, qPCR and RT-PCR (experiment design, reaction setup, isolation of genomic DNA, isolation of RNA).
  • RNA/DNA electrophoresis.
  • Isolation of protein, protein electrophoresis, and Western blotting.
  • Flow cytometry (design and conduct of an entire experiment).
  • Proactively seeking out new information in the literature and incorporating this into individual projects as well as the overall program.
  • Presenting research results and methodologies at weekly meetings with the immediate supervisor and project leader, project team meetings, and other research forums.
  • Mentoring and supervising junior team members by providing scientific expertise and sharing technical skills.
Your Background
  • PhD with preferably 2+ years of experience.
  • Prior industrial or relevant academic experience.
  • Previous experience in the fields of molecular biology.
  • Previous experience in the field of genome engineering (e.g. CRISPR/Cas9, TALEN, ZFN).
  • Experience in the in vitro eukaryotic cell line culture as well as in chemical transfection and viral transduction.
  • Experience in working with bacteria (transformation, amplification etc.).
  • Very good knowledge and experience in molecular cloning techniques (design and conduct of the entire procedure).
  • Experience in working with DNA, RNA, and proteins.
  • Technical expertise with standard molecular biology techniques like ELISA, Western Blot, qPCR, as well as cytotoxicity, proliferation, apoptotic, and cellular reporter system assays.
  • Expertise with flow cytometry, cytokine release, cytotoxicity and proliferation assays is strongly preferred.
  • Experience in working with cell lines and cell-based assays in microplate format (experience with 3D assays will be a plus).
  • Expertise in working in GxP environment is preferred.
  • Practical experience with analysis software such as FlowJo, GraphPad Prism, SnapGene, and Excel is strongly preferred.
  • Ability to manage large amounts of data and to supervise less experienced scientists/research assistants.
  • Willingness to learn and adopt new experimental techniques.
  • Active participation in task performance and problem-solving attitude.
  • Good command of spoken and written English.
